A new method for construction of Ti plasmid-less strains in Agrobacterium tumefaciens.
Agrobacterium tumefaciens harboring a Ti plasmid causes plant tumorigenesis and it's DNA transfer system have been extensively used in plant biotechnology. Because Ti plasmids are high stable in agrobacterial hosts, their curing is very difficult by conventional methods. Here, we introduced a novel curing method by using Ti plasmids incompatibility.

A wide host range cosmid vector has been constructed by insertion of the lambda cos site into the plasmid pRK2501. This cosmid, which is maintained in Agrobacterium tumefaciens and is compatible with the Ti plasmid, has been used to make a clone bank of the A. tumefaciens pTiA6 plasmid. Several pTiA6 cosmids have been used to complement Tn5-induced Ti plasmid mutations. Five avirulent mutations...
متن کاملUse of ti plasmid DNA probes for determining tumorigenicity of agrobacterium strains.
Probes consisting of T-DNA genes from the Ti plasmid of Agrobacterium tumefaciens were used for determining tumorigenicity of strains. Two P-labeled probes hybridized with 28 of 28 tumorigenic strains of the pathogen but not with 20 of 22 nontumorigenic strains. One probe, pTHE17, consists of all but the far left portion of the T-DNA of strain C58. Probe SmaI7 consists of SmaI fragment 7 of pTi...

gene can be spliced to the virus vector. Also, the added gene affects the cell-to-cell movement (invasiveness) of the virus, since the virus can no longer mature. Moreover, the virus is mainly limited to Cruciferae as host plants and therefore the prospects of infecting other major crops are narrow. Nevertheless, in-depth studies on this virus will help provide important information on means to...
متن کاملTi plasmid-controlled chromosome transfer in Agrobacterium tumefaciens.
In octopine-type A. tumefaciens R10, transfer of chromosomal arginine degradation genes (arc genes) was observed under conditions in which Ti plasmid transfer took place. However, transconjugants that had acquired the arc genes but not the Ti plasmid were recovered. During this process, several other chromosomal genes, such as genes encoding phage resistances or genes complementing a galactose ...
